分子生物學,生物化學,細胞生物學,遺傳學及表觀遺傳學,非編碼RNA (noncoding RNA),基因表達及調控,癌症生物學。
研究經驗和研究領域:
1)藥用植物抑癌活性成分的藥效機理研究:藥用植物中的很多成分具有抑癌活性,但其藥效機理不明,限制其臨床應用。為突破該瓶頸,我們将探究植物藥用成分在肝癌細胞内的分子靶點、所影響的重要基因及細胞生物途徑,創立科學的聯合給藥方式以有效抑制肝癌發展并降低對機體副作用。首次在細胞水平驗證了鞣花酸聯合阿黴素或聯合順鉑的抗肝癌活性,并在小鼠模型中鑒定了鞣花酸聯合阿黴素使用,可以顯著加強阿黴素抗肝癌活性,減弱由高濃度下阿黴素引發的心肌毒性。此外,我們還研究了百裡香琨,地榆皂苷II的植物天然活性物質的抑癌活性。
2)發明了以DNA載體為基礎的RNA幹擾(RNAi)技術(即shRNA方法)。這一技術的發明為許多生物學科的研究提供了全新的方法,尤其是對基因和蛋白質功能的研究具有重大的創新性,也進了RNAi技術在醫療上的應用。目前,這一方法已經成為世界範圍實驗室用于研究基因功能的常規實驗技術。該方法的DNA載體成為Ambion/Life Technologies公司的第一代産品(商品名pSilencer)。做為第一作者的相關PNAS文章已被引用上千次。
3)利用對Yin Yang 1(YY1)蛋白在腫瘤發生中功能的研究來探索幾種常見癌症(乳腺癌,前列腺癌,肺癌)的緻病機制。我的多年的研究成果确立了YY1的癌基因地位。作為一個多功能蛋白,YY1在癌症發生中的終極作用一直有争議。我在2004年發表的Cell文章中發現了YY1對p53的負調控,奠定了YY1的癌基因的地位。這些研究進一步确定YY1的癌基因地位,并揭示了YY1在癌症治療中作為靶基因的可能性。通過幹擾YY1與癌蛋白之間相互作用以抑制腫瘤生長的多肽已被授權專利。
4)對其它抑癌基因、原癌基因及非編碼RNA的研究:(1)利用基因芯片等方法對抑癌基因SOX7的研究,發現其上、下遊調控因子及可能的調節機制;(2)對DMTF1研究,發現了這一基因産生mRNA的不同選擇性剪接(alternative splicing)形态對乳腺癌發生和發展具有完全相反的作用,以及調節該可變剪接的分子機制;(3)研究了幾種microRNA對原癌基因EZH2、YY1和FASN的調節作用;(4)利用多個乳腺癌及它們對應的癌旁組織樣品中的長非編碼RNA(lncRNA)的分析,發現了多種在乳腺癌中差異性表達的lncRNA,目前正在對它們的生物學意義和對乳腺癌的調節功能進行鑒定。
